A win for England would take them a step closer to Russia 2016 as well as boost Gareth Southgate's chances of becoming manager on a full-time basis.
Defeat for Scotland would also push their own boss Gordon Strachan closer to the sack as he battles to keep alive his side's fading hopes of securing qualification.
There is no love lost between the oldest rivals in international football and it promises to be another no-nonsense showdown.
Kane admits he is relishing the battle and that England will be well pumped up come kick off.
The Tottenham striker said: "We want to go out there and win the game. We want to send the fans home happy, with a good result and good performance.
"We are definitely up for the game and we have to use all the positive energy from the home crowd.
"But we don't want to get any silly bookings or red cards. We want all our men out there on the pitch.
"There will be tackles flying in because that is the nature of the game, so it's important to keep a cool head and remains calm."
Kane admits he cannot stomach the prospect of losing to the Scots because of how painful the aftermath would be.
Kane, whose return from a long spell out with ankle ligament damage is a huge boost to Southgate, added:
"We are just concerned with winning this game. All we can do is our jobs on the pitch and put in a good performance.
"We know their players and we have Scottish friends who will certainly make it heard if they beat us."
