The strike was carried out in the western Libyan city of Sabratha at about 3.30am local time today, hitting a building in the Qasr Talil district.
An official said 41 people had been killed and six wounded, but the death toll could not be immediately verified.
Thirty-eight people, including 30 British tourists, were shot dead on a beach near the Tunisian city of Sousse in June, 2015.
The mastermind of last summer's Tunisia attacks in Sousse and at the Bardo Museum, Noureddine Chouchane, was a principal target of the strikes, and has reportedly been killed.
The Pentagon said: "Destruction of the camp and Chouchane's removal will eliminate an experienced facilitator and is expected to have an immediate impact on ISIL's ability to facilitate its activities in Libya, including recruiting new ISIL members, establishing bases in Libya, and potentially planning external attacks on US interests in the region."
US officials have said a senior ISIS operative is likely to have been killed in the bombing this morning.
The gunmen who carried out those attacks trained at camps in Libya.

US officials are reportedly trying to establish whether Chouchane has been killed in the attacks today.
Sabratha is near the Tunisian border, and is one of the areas where ISIS militants are expanding.
Recent reports say top ISIS commanders and many fighters have fled to Libya from Iraq and Syria because of intense bombing there.
