But Spurs boss Mauricio Pochettino insists he will be fit to return for their Premier League title charge at home to Swansea on Sunday.
Kane did not train with his team-mates yesterday ahead of their last 32, second-leg clash with the Italians as he was seeing a specialist.
And although Hugo Lloris will start after returning from a shoulder injury, there was more bad news for Tottenham with Mousa Dembele ruled out for at least 10 days after a groin injury.
Nacer Chadli is likely to start up front tonight for Spurs after scoring the away goal in their 1-1 draw in Florence last week.
But Dembele could miss their next four matches, including next Wednesdays trip to West Ham and the north London derby against Arsenal on March 5.
Pochettino, whose side hope to bounce back after their FA Cup defeat to Crystal Palace, said: “We are in a key period. It’s very important for us. We know that. We need to be ready at the right level.”
